Nah, that's not unusual for Singapore. Old colonial buildings and forgotten bits are everywhere. Probably just an old utility access point, maybe for pipes or wiring. Or maybe it leads to a secret speakeasy... that's way more fun to imagine, though. Doubt it's anything sinister, but if you're really curious, maybe grab a friend and check it out during the day. Better safe than sorry, especially around that big old banyan tree. They do get some pretty gnarly roots.
