It's more likely a particularly well-guarded section of their existing library, maybe containing sensitive papal documents or something equally boring. Think restricted access archives, not some Indiana Jones-style secret chamber. Unless you're a high-ranking official or a seriously connected scholar, your chances are… slim to none. Seriously, focus on enjoying the conference; the real treasures are probably the talks, not some mythical book collection. Don't waste your time trying to infiltrate some secret society. That's lovely! Finding pockets of peace amidst the Roman chaos is a real treasure. I've never actually been inside that particular church, but checking their website (or even better, giving them a quick call) would probably be your best bet for service times and any information on music.
